The subject invention concerns a method of introducing at least one DNA sequence expressing a protein or protein fragment which substantially alleviates articular cartilage defects. This methods involves in vitro culture of chondrocytes, transfection of the chondrocytes with a recombinant vector housing the DNA sequence to be expressed, and delivery of the transfected chondrocytes to the damaged cartilage region. This method can also be used in tandem with synovial cell delivery techniques of the present invention. This method is also useful as a model in animal studies regarding joint pathologies.
L'invention concerne un procédé d'introduction d'au moins une séquence d'ADN qui exprime une protéine ou un fragment de protéine qui atténue sensiblement les défauts cartilagineux d'articulations. Le procédé comprend la mise en culture in vitro de chondroblastes, la transfection des chondroblastes à l'aide d'un vecteur recombinant qui contient la séquence d'ADN à exprimer, et l'administration des chondroblastes transfectés à la région cartilagineuse atteinte. On peut également utiliser ce procédé avec les techniques décrites d'administration de cellules synoviales. Ce procédé est également utile comme modèle dans les études de pathologies articulaires animales.
