ABSTRACT Restriction endonuclease buffer compositions which are suitable for use with essentially all restriction endonucleases. The compositions comprise Tris-HCl, MgCl2, and at least one of KCl, beta-mercaptoethanol and deoxyribonucleoside triphosphate. The buffer compositions provide efficient restriction of DNA using a wide variety of restriction endonucleases, regardless of the specific buffer requirements of the restriction endonuclease when using conventional restriction buffers.
