Attempts have been made to develop host cells capable of producing glycoprotein compositions such as antibody compositions useful in drug discovery. It is intended to provide a cell having been neutralized to a serum- free medium and carrying a knockout genome gene of an enzyme which participates in a sugar chain modification whereby fucose is attached via an .alpha.-bond at the 1-position to the 6-position of N-acetylglucosamine at the reducing end of an N-glycoside-binding complex sugar chain; and a process for producing a glycoprotein composition with the use of this cell.

L'invention concerne un procédé permettant de développer des cellules hôtes capables de produire des compositions de glycoprotéines, telles que des compositions d'anticorps pouvant être utilisées dans la recherche sur les médicaments. En outre, cette invention concerne une cellule neutralisée jusqu'à obtention d'un support exempt de sérum et transportant un génome d'une enzyme impliquée dans une modification de la chaîne glucidique au cours de laquelle le fucose est relié par l'intermédiaire d'une liaison .alpha., au niveau de la position 1, à la position 6 de la N-acétylglucosamine à l'extrémité de réduction d'une chaîne glucidique complexe se liant au N-glycoside. Cette invention concerne également un procédé permettant de produire une composition de glycoprotéines au moyen de la cellule susmentionnée.
