The invention relates to the field of genetic engineering, more specifically to the field of production of glycoproteins, more specifically to the engineering of the glycosylation pattern of glycoproteins. Provided is a method for producing a glycoprotein in a recombinant host cell, comprising allowing expression of said glycoprotein in a basidiomycete host provided with N-acetylglucosamine : alpha-3-D-mannoside beta-1,2-N- acetylglucosaminyltransferase (GnT-I; EC 2.4.1.101) activity. Also provided is a basidiomycete host cell expressing GnT-I, preferably human GnT-I and use thereof for the manufacture of (mammalian) proteins carrying complex N- glycans, such as (monoclonal) antibodies.

L'invention se rapporte au domaine du génie génétique, particulièrement au domaine de production de glycoprotéines, plus particulièrement au domaine de l'ingénierie du motif de glycosylation de glycoprotéines. On décrit une méthode de production d'une glycoprotéine dans une cellule hôte recombinante, qui consiste à permettre l'expression de ladite glycoprotéine dans un hôte de basidiomycètes présentant une activité N-acétylglucosamine: alpha-3-D-mannoside bêta-1,2-N-acétylglucosaminyltransférase (GnT-I; EC 2.4.1.101). On décrit en outre une cellule hôte de basidiomycètes exprimant la GnT-I, de préférence la GnT-I humaine, et son utilisation pour la production de protéines (mammaliennes) contenant des glycans complexes, tels que des anticorps (monoclonaux).
