An object of the present invention is to provide an oligosaccharide chain added GLP-1 peptide that has higher stability in blood than that of GLP-1 and, preferably, exhibits higher activity of controlling blood-sugar levels than that of GLP-1. The present invention provides an oligosaccharide chain added GLP-1 peptide having GLP-1 activity, wherein at least two amino acids are each substituted with an oligosaccharide chain added amino acid, in (a) GLP-1; (b) a peptide having the amino acid sequence of GLP-1 with deletion, substitution or addition of one or several amino acids; or (c) a GLP-1 analog. The present invention also provides an oligosaccharide chain added GLP-1 peptide having GLP-1 activity, wherein at least one amino acid is substituted with an oligosaccharide chain added amino acid, in the (a), (b) or (c) and the oligosaccharide chain is an oligosaccharide chain comprising oligo hyaluronic acid. In the oligosaccharide chain added amino acid, the oligosaccharide chain may be linked to the amino acid via a linker.
La présente invention concerne un peptide GLP-1 glycosylé ayant une demi-vie plus longue dans le sang et de préférence une activité plus élevée pour abaisser le taux de glycémie par rapport à celles de GLP-1. La présente invention concerne spécifiquement un peptide GLP-1 glycosylé ayant une activité GLP-1, qui est caractérisé en ce qu'il comporte la substitution d'au moins deux résidus d'acide aminé dans le peptide suivant (a), (b) ou (c) par des résidus d'acide aminé glycosylés : (a) GLP-1 ; (b) un peptide ayant la délétion, la substitution ou l'addition d'un ou plusieurs résidus d'acide aminé dans GLP-1 ; ou (c) un analogue de GLP-1. La présente invention concerne en outre, spécifiquement, un peptide GLP-1 glycosylé ayant une activité GLP-1, qui comporte la substitution d'au moins un résidu d'acide aminé dans le peptide (a), (b) ou (c) par un résidu d'acide aminé glycosylé, où la chaîne glucidique comprend un acide oligohyaluronique. Dans le résidu d'acide aminé glycosylé, une chaîne glucidique et un résidu d'acide aminé peuvent être liés l'un à l'autre par l'intermédiaire d'un lieur.
