The present invention relates to a lipopolysaccharide moiety comprising a conserved triheptosyl inner-core moiety of lipopolysaccharide substantially free of variable outer core oligosaccharide chain extension, and to vaccines obtaines therefrom which are cross-reactive for Haemophilus influenzae strains. The invention also relates to defined mutations in the biosynthetic machinery for lipopolysaccharide (LPS) expression in Haemophilus influenzae useful to obtain the abovementioned moiety. The invention also relates to using conjugates of the LPS from the mutant strains so obtained to elicit a heterologous immune response against a wide range of disease-causing H. influenzae strains. More specifically, the invention relates to vaccines for prevention of bacterial infections comprising core lipopolysaccharide of Haemophilus influenzae. A lipopolysaccharide moiety comprising a conserved triheptosylinner-core moiety of lipopolysaccharide substantially free of variable outer core oligosaccharide chain extensions.

Cette invention se rapporte à une fraction de lipopolysaccharide comprenant une fraction noyau interne triheptosyle conservée de lipopolysaccharide, essentiellement exempte d'allongements de chaîne d'oligosaccharide à noyau externe variable, ainsi qu'à des vaccins obtenus à partir de cette fraction et qui entrent en réaction croisée avec des souches d'Haemophilus influenzae. Cette invention concerne également des mutations définies dans le processus de biosynthèse pour l'expression de lipopolysaccharide (LPS) dans Heamophilus influenzae, utilisée pour obtenir la fraction mentionnée ci-dessus. Cette invention concerne également l'utilisation de conjugués de LPS provenant des souches mutantes ainsi obtenues pour déclencher une réponse immunitaire hétérologue contre une large gamme de souches d'H. influenzae causant la maladie. Cette invention concerne plus spécifiquement des vaccins pour prévenir des infections bactériennes comprenant un lipopolysaccharide noyau d'Haemophilus influenzae. Cette invention a pour objet une fraction de lipopolysaccharide comprenant une fraction noyau interne triheptosyle conservée de lipopolysaccharide, essentiellement exempte d'allongements de chaîne d'oligosaccharide à noyau externe variable.
