The present invention relates to hepatitis B virus (hereinafter it refers to HBV) polymerase containing a histidine tag, RNase H enzyme derived from HBV polymerase and processes for preparation thereof. More particularly, the present invention relates to recombinant HBV polymerase, its RNase H domain with enzyme activity, expression vectors producing the enzymes in E. coli and processes for preparing the HBV polymerase and the RNase H enzyme which can be easily purified due to their histidine tags. The present invention relates to uses of the HBV polymerase and the RNase H enzyme for screening antiviral agents.

L'invention concerne la polymérase du virus de l'hépatite B (ci-après HBV) qui contient une étiquette d'histidine et un enzyme RNase H dérivé de la polymérase du virus de l'hépatite B, ainsi que des procédés de préparation. L'invention se rapporte notamment à une polymérase recombinante du HBV, à son domaine RNase H à activité enzymatique, à des vecteurs d'expression produisant les enzymes dans E. coli et à des procédés de préparation de la polymérase HBV et de l'enzyme RNase H, que l'on peut aisément purifier grâce à leurs étiquettes d'histidine. En dernier lieu, l'invention se rapporte à de différentes utilisations de la polymérase HBV et de l'enzyme RNase H dans le criblage d'agents antiviraux.
