The present invention provides a. hepatitis C virus gene, a replicon RNA derived from the gene, a replicon-replicating cell into which the replicon RNA is introduced, and a method for screening a drug using the replicon-replicating cell. By introducing a replicon RNA comprising (A) a polynucleotide having the nucleic acid sequence shown in SEQ ID NO: 5; (B) a polynucleotide having the nucleic acid sequence shown in SEQ ID NO: 7; (C) a polynucleotide coding for a polypeptide having the amino acid sequence shown in SEQ ID NO: 6; (D) a polynucleotide coding for a polypeptide having the amino acid sequence shown in SEQ ID NO: 8; or (E) a polynucleotide having a nucleic acid sequence having a homology of not less than 90% with the nucleic acid sequence shown in SEQ ID NO: 5 or SEQ ID NO: 7; or a replicon RNA of the genotype lb comprising nucleotides coding for the 1804th amino acid leucine and the 1966th amino acid lysine in the amino acid sequence of a hepatitis C virus polyprotein and a polynucleotide coding for an NS4B protein, into a cell, the replicon-replicating cell can be prepared. By using the replicon- replicating cell, the screening for the drug can be carried out.

L'invention a pour but de fournir un gène du virus de l'hépatite C, un réplicon ARN prenant naissance dans ce gène, une cellule à réplication autonome dans laquelle est transféré ce réplicon ARN et un procédé de criblage d'un médicament par l'utilisation de cette cellule à réplication autonome. Une cellule à réplication autonome peut être construite en transférant dans une cellule un réplicon ARN contenant : (A) un polynucléotide qui comprend la séquence de base représentée par la SEQ ID N° : 5 ; (B) un polynucléotide qui comprend la séquence de base représentée par la SEQ ID N° : 7 ; (C) un polynucléotide qui code un polypeptide comprenant la séquence d'acides aminés représentée par la SEQ ID N° : 6 ; (D) un polynucléotide qui code un polypeptide comprenant la séquence d'acides aminés représentée par la SEQ ID N° : 8 ; ou (E) un polynucléotide qui comprend une séquence de base ayant une homologie de 90 % ou plus avec la séquence de base représentée par la SEQ ID N° : 5 ou 7 ; ou un réplicon ARN de génotype 1b qui contient un nucléotide codant la leucine située en position 1804 et la lysine située en position 1996 dans la séquence d'acides aminés de la polyprotéine du virus de l'hépatite C et un polynucléotide codant la protéine NS4B. L'utilisation de cette cellule à réplication autonome rend possible le criblage d'un médicament.
