The present invention features methods for enhancing the ability of a genotype 2b NS5B sequence to function in a replicon, for producing replicons containing a functional genotype 2b NS5B, and for using replicons to measure the ability of a compound to affect HCV replication that is sustained with the genotype 2b polymerase. Also featured is a genotype 1b NS4B adaptive mutation. The ability to produce replicons containing a functional genotype 2b NS5B is illustrated by the production of chimeric replicons based on HCV genotype 1b where substantially all the NS5B sequence is replaced with a genotype 2b NS5B.
L'invention concerne des procédés permettant d'accroître la capacité d'une séquence de la NS5B du génotype 2b de fonctionner dans un réplicon, aux fins de production de réplicons renfermant une NS5B fonctionnelle du génotype 2b et aux fins d'utilisation des réplicons dans la mesure de la capacité d'un composé d'avoir un effet sur la réplication du VHC, supportée au moyen de la polymérase du génotype 2b. L'invention concerne également une mutation adaptative de la NS4B du génotype 1b. La capacité de produire des réplicons renfermant une NS5B fonctionnelle du génotype 2b est illustrée par la production de réplicons chimériques en fonction du génotype 1b du VHC où sensiblement l'ensemble de la séquence de la NS5B est remplacée par une NS5B du génotype 2b.
