The present invention relates to methods of reducing heme loss in hemoglobins to produce stability and improve expression yield of hemoglobins, particularly recombinant hemoglobins. Such methods are accomplished by introducing mutations in the alpha or beta subunits of hemoglobins to increase heme affinity. The present invention further relates to novel mutations that reduce such heme loss.

La présente invention se rapporte à des procédés permettant de diminuer la perte d'hèmes dans des hémoglobines dans le but de stabiliser ces hémoglobines et d'accroître leur expression, et ce tout particulièrement dans des hémoglobines recombinées. Ces procédés consistent à introduire des mutations dans les sous-unités alpha ou bêta des hémoglobines de façon à accroître leur affinité pour les hèmes. La présente invention se rapporte en outre à de nouvelles mutations qui réduisent la perte d'hèmes.
