Hemoglobin conjugates useful as hemoglobin-based oxygen carriers are prepared by reacting hemoglobin with oxidatively ring-opened polysaccharides such as hydroxyethyl starch or dextran, and storing the resultant conjugate under conditions which allow it to transform to a lower molecular weight product, after conjugation. The conjugate is then reductively stabilized to form secondary amino bonds between the hemoglobin and the polysaccharide, and formulated as a HBOC.

La présente invention concerne la préparation de conjugués de l'hémoglobine utiles en tant que transporteurs d'oxygène à base d'hémoglobine. A cette fin, on fait réagir de l'hémoglobine avec des polysaccharides à cycle ouvert par oxydation tels que de l'amidon hydroxyéthylée ou de la dextrane, le conjugué qui en résulte étant conservé dans des conditions qui lui permettent de se transformer, après conjugaison, en un produit à faible poids moléculaire. Le conjugué est ensuite stabilisé réductivement pour former des liaisons amino entre l'hémoglobine et le polysaccharide, et formulé en tant que transporteur d'oxygène à base d'hémoglobine.
