Herpesviral particle preparations, e.g. a preparation of herpesviral particles isolated from the cell culture in which such particles were produced, can have at least part of the VP22 tegument protein present in the form of a recombinant mutant form of VP22, e.g. as a recombinant fusion polypeptide comprising a VP22-active sequence and a non-VP22 peptide or polypeptide sequence such as a fluorescent GFP sequence: corresponding DNA preparations are described. The use of virus particles containing fluorescent fusion protein to detect the progress of cell infection by virus and to screen for neutralising antibody or inhibitors of infection is also described. Vaccine uses of modified herpesvirus particles are described.

Selon cette invention, des préparations de particules d'herpèsvirus, p.ex., une préparation de particules d'herpèsvirus isolées à partir d'une culture de cellules dans laquelle ces particules ont été produites, possèdent au moins une partie de la protéine de tégument VP22, présente sous la forme d'un mutant recombinant de VP22, c'est-à-dire d'un polypeptide de fusion recombinant qui comprend une séquence active de VP22 et une séquence de peptide ou de polypeptide qui ne vient pas de VP22, par exemple, une séquence fluorescente de GFP. L'invention concerne aussi des préparations d'ADN correspondantes, l'utilisation de particules virales contenant la protéine de fusion fluorescente pour détecter l'évolution de l'infection d'une cellule par un virus et pour l'examen d'anticorps neutralisants ou d'inhibiteurs de l'infection. Elle concerne enfin l'utilisation des particules d'herpèsvirus modifié dans les vaccins.
