Herpesviruses naturally deficient in gD, that is to say not expressing their gD gene in vitro or not possessing this gene, are transformed to express gD in vitro. This transformation by genetic recombination can consist in replacement of the natural promoter by another one or insertion of an expression cassette containing gD and a promoter. The invention also relates to these viruses incorporating a gene coding for an antigen of interest, to the vaccines obtained and to the methods of culture thus improved. The invention relates in particular to avian herpesviruses, especially MDV, and the VZV virus.
Les herpèsvirus naturellement déficients en gD, c'est-à-dire n'exprimant pas leur gène gD in vitro ou ne possédant pas ce gène, sont transformés pour exprimer gD in vitro. Cette transformation par recombinaison génétique peut consister dans le remplacement du promoteur naturel par un autre ou l'insertion d'une cassette d'expression contenant gD et un promoteur. L'invention concerne aussi ces virus incorporant un gène codant pour un antigène d'intérêt, les vaccins obtenus et les procédés de culture ainsi améliorés. L'invention concerne notamment les herpèsvirus aviaires, en particulier MDV, et le virus VZV.
