Therapeutically active, heteroaryl-substituted deoxy glycero- phosphoethanolamines are disclosed having general formula (I) wherein one of X, Y or Z is a fatty ether substituent, one is a heteroaryl ring substituent with 1 - 4 nitrogens as the only heteroatoms, one of which is bonded to a carbon of the glyceryl backbone and one is a phosphoethanolamine substituted at the nitrogen, provided that each of X, Y and Z is a different substituent. Also disclosed are intermediate in processes for making compounds of formula (I). The intermediates are represented by a modification of general formula (I) in which the phosphoethanolamine substituent is replaced with an hydroxyl group. The invention encompasses all optical isomers and stereoisomers of the compounds of formula (I) and the intermediates and salts of these compounds, including the isomers. The therapeutically active compounds of the invention have anti-tumor, anti-psoriatic and anti-inflammatory activities. The invention entails pharmaceutical compositions comprising the therapeutically active compounds and methods of using the therapeutically active compounds to treat cancerous tumors, psoriasis and inflammation.
L'invention concerne des désoxy glycéro-phosphoéthanolamines substituées par hétéroaryle, actives sur le plan thérapeutique, qui ont la formule générale (I) dans laquelle parmi X, Y et Z, un désigne un substituant d'éther gras, un désigne un substituant de composé cyclique hétéroaryle ayant entre 1 - 4 azotes comme seuls hétéroatomes, dont un est lié à un carbone du squelette glycéryle, et un est un substituant de phosphoéthanolamine, substitué au niveau de l'azote, sous réserve que chacun de X, Y et Z soit un substituant différent. L'invention concerne également des intermédiaires utilisés dans des procédés servant à préparer des composés de la formule (I). Les intermédiaires sont représentés par une modification de la formule générale (I) dans laquelle le substituant de la phosphoéthanolamine est remplacé par un groupe hydroxyle. L'invention inclut tous les isomères et les stéréoisomères optiques des composés de la formule (I) et les intermédiaires et les sels de ces composés, y compris les isomères. Ces composés actifs sur le plan thérapeutique, ont des effets anti-psoriatiques et anti-inflammatoires. L'invention comprend des compositions pharmaceutiques contenant ces composés actifs sur le plan thérapeutique, ainsi que des procédés d'utilisation de ces composés à effet thérapeutique pour traiter des tumeurs cancéreuses, le psoriasis et les phénomènes inflammatoires.
