The invention is based on the discovery that recombinagenic oligonucleobases are active in prokaryotic cells that contain a strand transfer activity (RecA) and mismatch repair activity (MutS). Using this system a type of Duplex Mutational Vector termed a Heteroduplex Mutational Vector, was shown to be more active in prokaryotic cells than the types of mutational vectors heretofore tested. Further improvements in activity were obtained by replacing the tetrathymidine linker by a nuclease resistant oligonucleotide, such as tetra-2'-O-methyl-uridine, to link the two strands of the recombinagenic oligonucleobase and removing the DNA-containing intervening segment. The claims concern Duplex Mutational Vectors that contain the above improvements. In an alternative embodiment the claims concern the use of Duplex Mutational Vectors in prokaryotic cells.
L'invention concerne la découverte du rôle actif des oligonucléobases de recombinaison dans les cellules eucaryotes qui présentent une activité de transfert de brin (RecA) et une activité de réparation de mésappariement (MutS). Sur la base d'un tel système, on a montré qu'un vecteur duplex mutationnel appelé vecteur hétéroduplex mutationnel était plus actif dans les cellules procaryotes que les types de vecteurs mutationnels testés jusqu'à présent. On a encore amélioré l'activité en remplaçant le segment de liaison tétrathymidine par un oligonucléotide résistant à la nucléase, du type tétra-2'-O-méthyl-uridine, de manière à lier les deux brins de l'oligonucléobase de recombinaison. Une autre amélioration a consisté à éliminer le segment intervenant qui contient de l'ADN. L'invention concerne donc des vecteurs hétéroduplex mutationnels présentant les améliorations décrites. Selon un aspect, l'invention concerne l'utilisation desdits vecteurs dans les cellules procaryotes.
