The present invention provides a protein having high- affinity choline transporter activity which is important physiologically, a gene encoding the protein, and a method of screening a material promoting the high-affinity choline transporter activity with the use of the same, and the like. By examining high-affinity choline uptake activity of Na+- dependent transporter cDNA deduced from the genomic sequence of a nematode (C. elegans) in a Xenopus oocyte expression system, the cDNA (cho-1) of nematode high-affinity choline transporter is identified. Then the cDNA (CHT1) of rat high-affinity choline transporter is cloned from rat spinal cord by using the homology of a base sequence to this cDNA as an index. Similarly, the cDNA of human high-affinity choline transporter is cloned from human genome.

L'invention porte: sur une protéine physiologiquement importante présentant une forte activité de transporteur de choline à forte affinité; sur un gène codant pour ladite protéine; et sur un procédé de criblage d'une substance favorisant l'activité dudit transporteur de choline et son utilisation, etc. L'activité sur l'absorption de la choline de l'ADNc du transporteur de choline dépendant du Na<+>, qui se déduit de la séquence génomique d'un nématode<i>C.elegans</i>, s'examine dans un système d'expression de l'oocyte Xenopus laevis pouvoir identifier l'ADNc (cho-1) du transporteur de choline du nématode. En utilisant comme indication l'homologie d'une séquence de base avec cet ADNc, on peut cloner l'ADNc (CHT1) du transporteur de choline du rat à partir de sa moelle épinière. On peut de même cloner le transporteur de choline de l'homme à partir du génome humain.
