A process for obtaining a spray-dried high amylose sodium carboxymethyl starch comprising a major fraction of amorphous form and optionally a minor fraction of crystalline V form, is provided. The process comprises providing an amorphous pregelatinized high amylose sodium carboxymethyl starch (HASCA); dispersing the amorphous pregelatinized HASCA in a solution comprising water and at least one first pharmaceutically acceptable organic solvent miscible with water and suitable for spray-drying; and spray-drying the dispersion to obtain the spray-dried HASCA comprising a major fraction of amorphous form and optionally a minor fraction of crystalline V form, in the form of a powder. Also provided is a spray-dried HASCA sustained-release excipient. This excipient is useful for preparing a tablet for the sustained-release of at least one drug.
La présente invention concerne un procédé permettant d'obtenir de l'amidon carboxyméthylé au carboxyméthyle de sodium à teneur élevée en amylose desséché par pulvérisation comportant une fraction majeure de forme amorphe et éventuellement une fraction mineure de forme cristalline V. Le procédé comprend la mise à disposition d'un amidon carboxyméthylé au carboxyméthyle de sodium, et à contenu élevé d'amylose amorphe prégélatinisé (HASCA); la dispersion de l'HASCA amorphe prégélatinisé dans une solution comprenant de l'eau et au moins un premier solvant organique pharmaceutiquement acceptable miscible avec l'eau et apte à être soumise à un séchage pulvérisation; et le séchage par pulvérisation de la dispersion pour obtenir l'HASCA séché par la pulvérisation comportant une fraction majeure de forme amorphe et éventuellement une fraction mineure de forme cristalline V, sous forme d'une poudre. L'invention concerne également un excipient à libération prolongée à base d'HASCA séché par pulvérisation. Cet excipient est utile pour préparer un comprimé pour la libération prolongée d'au moins un médicament.
