A method for producing rAAV includes a first step of co-infecting a producer cell with a first recombinant herpes simplex virus containing AAV rep and cap genes, and a second recombinant herpes simplex virus containing a promoter and transgene flanked by two AAV ITRs; a second step of culturing the co-infected producer cell under conditions that result in the production of rAAV in the cell; and a third step of collecting the rAAV produced.

L'invention concerne un procédé relatif à la production de rAAV, selon trois étapes: co-infection de cellule productrice avec un premier virus d'herpès simplex de recombinaison renfermant des gènes rep et cap d'AAV, et avec un second virus d'herpès simplex de recombinaison renfermant un promoteur et un transgène que flanquent deux répétitions terminales inversées d'AAV; mise en culture de la cellule productrice co-infectée, dans des conditions induisant la production de rAAV à l'intérieur de la cellule; et collecte du rAAV résultant.
