This invention is directed to a continuous process for producing viral particles comprising providing in a perfused growth medium a population of viable virally infected non-lytic cells, and removing medium containing said cells at a rate to maintain the steady-state log-phase growth of cells remaining in said perfused growth medium. The invention is also directed to a process for purifying retroviral particles comprising passing a solution comprising the retroviral particles and contaminants through an anion exchange resin, and eluting the retroviral particles from the resin.
L'invention concerne un procédé permettant de produire en continu des particules virales et consistant à introduire une population de cellules viables non lytiques, infectées par un virus, dans un milieu de croissance de perfusion, puis à retirer le milieu contenant ces cellules à une cadence permettant de maintenir la phase de croissance exponentielle en régime permanent pour les cellules qui restent dans ledit milieu de croissance. L'invention concerne aussi un procédé permettant de purifier des particules rétrovirales et consistant à faire passer une solution qui contient ces particules et des contaminants au travers d'une résine d'échange anionique, puis à éluer ces particules rétrovirales de la résine.
