ABSTRACT Highly Sensitive Optical Immunoassay Using Enzyme-Labeled Reagents A thin film optical immunoassay device comprising a solid support substrate having an upper and a lower surface, and supporting on its upper surface, an unlabeled ligand antibody layer bound to said substrate, at least one layer comprising an immobilized enzyme conjugate, complexed with an analyte of interest and capable of further interacting with an enzyme reactive delivery substance to form an insoluble reaction product, said enzyme conjugate layer and said unlabeled antibody layer having a measurably increased mass change, said mass capable of precipitation by a precipitating agent applied as a substrate thereover. The invention also provides a corresponding process for detecting an analyte in a medium, and a diagnostic test kit for a thin film optical immunoassay.
