The present invention relates to a method for modifying the genome of eukaryotic cells by homologous recombination using DNA sequences which substantially differ from the target locus with respect to the nucleotide sequence (0.1 to 30 % divergence) in the region where recombination takes place. Homologous recombination between diverged sequences is achieved by genetic or transitory inactivation of the cell's mismatch repair system.

La présente invention se rapporte à un procédé de modification du génome de cellules eucaryotes par recombinaison homologue de séquences d'ADN qui diffèrent sensiblement du locus cible par rapport à la séquence nucléotidique (0,1 à 30 % de divergence) dans la région où se produit la recombinaison. On parvient à une recombinaison homologue entre des séquences divergentes par inactivation génétique ou transitoire du système de réparation des mésappariements des cellules.
