An inhibitor of platelet aggregation and adhesion is purified and characterized from soluble protein extracts of adult Ancylostoma caninum hookworms, and then cloned and sequenced. The inhibitor blocks platelet aggregation in response to a variety of agonists, interfering with the binding of at least one cell surface integrin with its respective ligand. Embodiments include inhibition of the binding of fibrinogen to cell surface integrin GPIIb/IIIa (.alpha.IIb.beta.3) and inhibition of the binding of collagen to cell surface integrin GPIa/IIa (.alpha.2.beta.1). Medical and veterinary pharmaceutical and immunological compositions containing the platelet inhibitor, and methods of using it, are described.

L'invention concerne un inhibiteur d'agrégation et d'adhésion plaquettaires purifié et caractérisé à partir d'extraits de protéine solubles d'ankylostomes adultes Ancylostoma caninum puis clonés et séquencés. Cet inhibiteur bloque l'agrégation plaquettaire en réponse à une grande variété d'agonistes, interférant avec la liaison d'au moins une intégrine en surface cellulaire avec son ligand. Selon des variantes, on prévoit l'inhibition de la liaison de fibrinogène avec l'intégrine en surface cellulaire GPIIb/IIIa (.alpha.¿IIb?.beta.¿3?) et l'inhibition de la liaison de collagène avec l'intégrine de surface cellulaire GPIa/IIa (.alpha.¿2?.beta.¿1?).
