Human and mouse TREK-1 potassium transport proteins are disclosed as well as a method for using potassium transport proteins for identifying substances having anesthetic properties, such as producing a safe, reversible state of unconsciousness with concurrent amnesia and analgesia in a mammal upon inhalation, in a method comprising (a) contacting the test substance with a mammalian potassium transport protein, wherein said potassium transport protein exhibits outward-going potassium rectification; and (b) determining the potassium transport activity of the potassium transport protein, wherein an activation of potassium transport is indicative of the test substance having anesthetic properties.

L'invention concerne des protéines de transport de potassium humaines et murines TREK-1 ainsi qu'un procédé d'utilisation de protéines de transport de potassium pour identifier les substances possédant des propriétés anesthésiques, par exemple, capables d'induire chez un mammifère les ayant inhalés un état d'inconscience sans danger et réversible avec l'amnésie et l'analgésie concomitantes. Le procédé consiste (a) à mettre en contant la substance de test avec une protéine mammalienne de transport de potassium, lesdites protéines de transport de potassium manifestant une rectification de potassium dirigé vers l'extérieur; (b) et à déterminer l'activité de transport de potassium de la protéine de transport de potassium, une activation de transport de potassium indiquant la substance test possédant des propriétés anesthésiques.
