A modified human complement C3 protein (C3) is disclosed comprising a substitution of a portion of a human C3 protein, with a corresponding portion of a Cobra Venom Factor protein (CVF) which results in a human C3 protein with CVF functions, but with substantially reduced immunogenicity. Advantageously, the C3 protein can be manipulated to contain at least one of the following CVF functions: increased stability of the C3 convertase and increased resistance to the actions of factors H and/or I. A large number of hybrid C3 proteins containing substitutions in the C-terminal portion of the alpha chain of C3 are presented and tested for the above functions. Methods of treatment of diseases such as reperfusion injury, autoimmune diseases, and other diseases of increased complement activation are presented as well as methods of increasing the effectiveness of gene therapeutics and other therapeutics.
L'invention concerne une protéine C3 du complément humaine modifiée qui comprend une substitution d'une partie d'une protéine C3 du complément humaine, et une partie correspondante d'une protéine du facteur du venin de cobra (CVF) qui permet d'obtenir une protéine C3 du complément humaine à fonctions CVF, mais dont l'immunogénicité est sensiblement réduite. De manière avantageuse, la protéine C3 du complément peut être manipulée de façon à contenir au moins une des fonctions CVF suivantes : stabilité améliorée de la convertase C3 et résistance améliorée aux actions des facteurs H et/ou I. Un grand nombre de protéines C3 hybrides contenant des substitutions dans la partie d'extrémité C de la chaîne alpha de C3 sont présentées et testées pour les fonctions précédemment mentionnées. L'invention concerne également des procédés de traitement de maladies de type lésion de reperfusion, maladies auto-immunes, et autres maladies d'activation de complément améliorée sont présentées ainsi que de procédés d'amélioration de l'efficacité de thérapies géniques et d'autres thérapies.
