A new form of human glandular kallikrein-1 (hK2) has been found by cloning a prepro-hK2 cDNA from a human prostate cancer tissue cDNA library. This form of hK2 differs in sequence from the previously deduced sequence for hK2 in that Arg226 is substituted by Trp. Use of a baculovirus expression vector is described for obtaining hK2 proteins, including Arg226-hK2 as an active mature protein directly from a culture medium.

L'invention concerne une nouvelle forme de kallicréine glandulaire humaine du type 1 (hK2) qui a été découverte par clonage d'un ADNc codant pour la prépro-hK2, à partir d'un collection d'ADNc de tissu cancéreux de la prostate humaine. Cette forme de hK2 diffère de la séquence précédente établie pour hK2 en ce sens que l'arginine en position 226 est remplacée par Trp. On décrit l'utilisation d'un vecteur d'expression d'un baculovirus pour l'obtention de protéines hK2, en particulier d'Arg?226¿-hK2, c'est-à-dire de la protéine active mûre, directement à partir d'un milieu de culture.
