The present invention relates to a method for producing a functional human immunoglobulin, wherein a human heavy chain immunoglobulin, devoid of any light chain, is expressed, comprising the steps of: a) transforming a filamentous host cell with a recombinant construct encoding a modified human heavy chain immunoglobulin, wherein the modifications comprise one or more mutations in the region of the heavy chain protein involved in contact with the light chain; b) culturing said filamentous host cell under conditions promoting expression of said modified human heavy chain immunoglobulin; and c) recovering said modified human heavy chain immunoglobulin.
La présente invention concerne un procédé de production d'une immunoglobuline humaine fonctionnelle, selon lequel une immunoglobuline humaine à chaînes lourdes, dépourvue de chaîne légère, est exprimée. Ce procédé consiste à : a) transformer une cellule hôte filamenteuse au moyen d'une construction recombinante codant pour une immunoglobuline humaine à chaînes lourdes modifiée, les modifications comprenant une ou plusieurs mutations dans la région de la protéine à chaînes lourdes impliquée se trouvant en contact avec la chaîne légère ; b) cultiver ladite cellule hôte filamenteuse dans des conditions favorisant l'expression de ladite immunoglobuline humaine à chaînes lourdes modifiée ; et c) récupérer ladite immunoglobuline humaine à chaînes lourdes modifiée.
