This invention relates to antibody engineering technology. More particularly, the present invention relates to human IgM antibodies and derivatives thereof, which have novel binding specificity with regard to several oligosaccharide sequences and/or xenoantigenic sialic aicd residue. The present invention also relates to processes for making and engineering such novel saccharide and/or NeuGc-binding monoclonal antibodies and to methods for using these antibodies and derivatives thereof in the field of immunodiagnostics, enabling qualitative and quantitative determination of xenoantigenic NeuGc in biological and raw material samples, as well as in immunotherapy, enabling blocking of xenoantigenic NeuGc in patients.
L'invention concerne une technologie d'ingénierie d'anticorps. L'invention concerne plus particulièrement des anticorps IgM humains et leurs dérivés, qui présentent une nouvelle spécificité de liaison par rapport à plusieurs séquences d'oligosaccharides et/ou un résidu d'acide sialique xénoantigénique. L'invention concerne également des procédés de fabrication et d'ingénierie de ces nouveaux anticorps monoclonaux se liant à NeuGc et/ou à des saccharides, ainsi que des méthodes d'utilisation de ces anticorps et de leurs dérivés dans le diagnostic immunologique, de manière à détecter de façon qualitative et quantitative NeuGc xénoantigénique dans des échantillons biologiques et de matière première, ainsi que dans l'immunothérapie, de manière à bloquer NeuGc xénoantigénique chez des patients.
