A human-type anti-blood coagulation factor VIII (hereinafter referred to as FVIII) which binds to human FVIII and specifically inhibits the blood coagulation activity thereof, and its antibody fragment. An scFv display phage library, which has been constructed by using scFv DNA on the basis of random combinations of the VH chain gene and VL chain gene of immunoglobulin originating in lymphocytes of a patient with hemophilia A, is reacted with FVIII having been immobilized on a solid phase via an anti-FVIII monoclonal antibody. Then scFv clones capable of binding to FVIII are screened to thereby clarify antibody VH chain and VL chain specific to FVIII.

Cette invention se rapporte à un anticorps contre le facteur VIII de coagulation sanguine (appelé ici FVIII) de type humain, qui se fixe au facteur FVIII humain et inhibe spécifiquement son activité de coagulation sanguine, et à son fragment d'anticorps. A cet effet, une bibliothèque de phages d'expression scFv, qui a été construite à l'aide d'un ADN scFv sur la base de combinaisons aléatoire du gène de chaîne VH et du gène de chaîne VL de l'immunoglobuline provenant des lymphocytes d'un patient atteint d'hémophilie A, est mise en réaction avec le facteur FVIII ayant été préalablement immobilisé sur un phase solide via un anticorps monoclonal anti-FVIII. Les clones scFv capables de se fixer au facteur FVIII sont ensuite criblés pour permettre de clarifier la chaîne VH et la chaîne VL de l'anticorps, spécifiques du facteur FVIII.
