A humanized antibody against tissue factor (TF) which comprises: A. a humanized H chain containing (1) an H chain V region containing the H chain CDR of a mouse monoclonal antibody against TF and the H chain FR of a human antibody, and (2) the H chain C region of a human antibody; and B. a humanized L chain containing (1) an L chain V region containing the L chain CDR of a mouse monoclonal antibody against TF and the L chain FR of a human antibody, and (2) the L chain C region of a human antibody. The mouse monoclonal antibody CDR is grafted into the human antibody to construct the humanized V region. Next, the FR thereof is replaced by the corresponding FR of another human antibody with a high homology, thus detecting a highly active humanized antibody.
L'invention porte sur un anticorps humanisé contre le facteur tissulaire humain, cet anticorps comprenant: A) une chaîne H humanisée contenant (1) une région V contenant la région déterminant la complémentarité (CDR) de la chaîne H d'un anticorps monoclonal de souris contre TF et la région du squelette (FR) de la chaîne H d'un anticorps humain, et (2) la région C de la chaîne H d'un anticorps humain; et B) une chaîne L humanisée contenant (1) une région V contenant CDR de la chaîne L d'un anticorps monoclonal de souris contre TF et la région du squelette (FR) de la chaîne L d'un anticorps humain, et (2) la région C de la chaîne L d'un anticorps humain. La région déterminant la complémentarité (CDR) de l'anticorps monoclonal de la souris est greffée dans l'anticorps humain pour former la région V humanisée. La région du squelette (FR) de cet anticorps est ensuite remplacée par la région du squelette (FR) correspondante d'un autre anticorps humain à très forte homologie, ce qui permet de détecter un anticorps humanisé extrêmement actif.
