The present invention is directed to humanized antibodies that specifically bind the protein 4-1BB. The antibodies can be made by grafting of the complementarity determining regions (CDR's) of mouse monoclonal antibody to human 4-1BB to the remaining portions of a human antibody and by making further amino acid replacements. In addition, a pharmaceutical composition that includes the humanized antibody can be made and can be used to treat autoimmune diseases to suppress an immune response. The humanized antibody of the invention has high affinity for human 4-1BB, and exhibits sequence similarity to human antibody. As a result, the pharmaceutical composition of the present invention can be used to treat autoimmune disease and act as an immunosuppressant in humans without much side-effect.
La présente invention porte sur des anticorps humanisés qui se lient spécifiquement à la protéine 4-1BB. Il est possible de produire les anticorps en greffant des régions déterminant la complémentarité de l'anticorps monoclonal de la souris contre la protéine humaine 4-1BB sur les autres parties d'un anticorps humain et en effectuant des remplacements d'autres acides aminés. Il est également possible de produire et d'utiliser une composition pharmaceutique comprenant l'anticorps humanisé pour traiter des maladies auto-immunes afin de supprimer une réponse immune. L'anticorps humanisé de cette invention présente une haute affinité pour la protéine humaine 4-1BB et présente également une similarité de séquence avec l'anticorps humain. En conséquence, il est possible d'utiliser la composition pharmaceutique de cette invention pour traiter les maladies auto-immunes, cette composition pouvant agir comme immunosuppresseur chez l'homme avec peu d'effets secondaires.
