The present invention describes the preparation and use of biologically and immunologically active humanized monoclonal antibodies to Shiga toxin, a toxin associated with HC and the potentially life-threatening sequela HUS transmitted by strains of pathogenic bacteria. The present invention describes how these humanized antibodies may be used in the treatment or prevention of Shiga toxin induced diseases. One aspect of the invention is the humanized monoclonal antibody which binds Shiga toxin where the constant regions are igG1-kappa and the variable regions are murine in origin. Yet another aspect of the invention is expression vectors and host cells transformed with such vectors which express the humanized monoclonal antibodies of the present invention.
L'invention porte sur la préparation et l'utilisation d'anticorps monoclonaux humanisés et à activité immunologique des toxines de Shiga, toxines associées au HC (colite hémorragique), et sur les séquelles de HUS (syndrome de Gasser) potentiellement thanatogènes transmises par des souches de bactéries pathogènes. L'invention décrit comment ces anticorps humanisés peuvent servir à traiter ou à prévenir des maladies induites par les toxines de Shiga. L'un des aspects de l'invention porte sur un anticorps monoclonal humanisé se fixant à la toxine de Shiga, dont les régions constantes sont des IgG1-kappa, et dont les régions variables sont d'origine murine. Un autre aspect porte sur des vecteurs d'expression et les cellules hôtes transformées par lesdits vecteurs qui expriment les anticorps monoclonaux humanisés de l'invention.
