It is described a hybrid molecule of for- mula GM-CSF-L-EPO or EPO-L-GM-CSF use- ful for the stimulation of hematopoiesis, com- prising GM-CSF and EPO molecules fused to- gether by means of a linker L having a vari- able length of amino acids, GM-CSF and EPO molecules being either complete molecules or fragments thereof. Such hybrid molecules ex- hibit a higher specificity of action on ery- throid differentiation if compared to that of an equimolar mixture of not-linked GM-CSF and EPO molecules. Further, the present in- vention describes: DNA sequences encoding such hybrid molecules, which stimulate the hematopoiesis; plasmid vectors containing the genes encoding hybrid molecules and direct- ing their synthesis in host cells; the use of such hybrid molecules for the preparation of a pharmaceutical composition and such pharma- ceutical composition useful for stimulation of hematopoiesis.
L'invention se rapporte à une molécule hybride de formule GM-CSF-L-EPO ou EPO-L-GM-CSF utilisée dans la stimulation de l'hématopoïèse, cette molécule comprenant des molécules GM-CSF et EPO fusionné entre elles au moyen d'un segment de liaison L ayant une longueur variable d'acides aminés, les molécules GM-CSF et EPO étant soit des molécules complètes, soit des fragments. Ces molécules hybrides présentent une spécificité d'action supérieure sur la différentiation érythroïde par rapport à celle d'un mélange équimolaire de molécules GM-CSF et EPO non liées. L'invention se rapporte en outre à des séquences d'ADN codant ces molécules hybrides qui stimulent l'hématopoïèse; à des vecteurs de plasmide contenant les gènes codant les molécules hybrides et dirigeant leur synthèse dans des molécules hôtes; à l'utilisation de ces molécules hybrides dans la préparation d'une composition pharmaceutique, et à cette composition pharmaceutique utilisée dans la stimulation de l'hématopoïèse.
