The present invention provides an MHC Class II antigen presentation enhancing hybrid polypeptide. The hybrid has an N-terminus comprising the mammalian Ii key peptide LMRKLPKPPKPVSKMR (SEQ ID NO:1) and modifications thereof which retain antigen enhancing activity, a C-terminus comprising an antigenic epitope in the form of a polypeptide or peptidomimetic structure which binds to the antigenic peptide bending site of an MHC Class II molecule, and an intervening chemical structure covalently linking the N-terminal and C- terminal components. Methods for identifying a molecule which functions within the context of an MHC class II antigen presentation enhancing hybrid in an equivalent fashion as the Ii key peptide are also described. The present invention also provides a metod for enhancing presentation of an MHC Class II restricted antigenic epitope to a T cell. Both in vivo and ex vivo methods are provided. A method for identifying a compound which inhibits MHC Class II antigen presentation is also provided.
La présente invention concerne un polypeptide hybride améliorant la présentation d'antigène de classe II du complexe majeur d'histocompatibilité (CMH). Cet hybride possède une terminaison N comprenant le peptide mammalien clé Ii LMRKLPKPPKPVSKMR (SEQ ID NO:1), et des modifications de ce peptide, qui retient une activité intensificatrice d'antigène, une terminaison C comprenant un épitope antigénique sous la forme d'un polypeptide ou d'une structure peptidomimétique qui se lie au site de courbure du peptide antigénique d'une molécule de classe II du CMH, ainsi qu'une structure chimique d'intercalation liant les composants des terminaisons C et N de manière covalente. L'invention concerne aussi des méthodes d'identification d'une molécule qui fonctionne, dans le contexte d'un hybride améliorant la présentation d'un antigène de classe II du CMH, d'une façon équivalente au peptide clé Ii. Elle concerne encore une méthode destinée à améliorer la présentation d'un épitope antigénique de classe II du CMH, à stimulation restreinte, à un lymphocyte T. Les méthodes, à la fois in vivo et ex vivo, sont présentées. L'invention concerne, en outre, une méthode destinée à identifier un composé qui inhibe la présentation d'un antigène de classe II du CMH.
