The present invention provides a method for preparing a spray-dried, compressible granular formulation for preparing pharmaceutical tablets in which essentially water-insoluble, acidic, amphoteric, and basic pharmaceutically active agents are converted to more water-soluble salts which are granulated with hydrolyzed cellulose, drug-containing slurries, the resulting granulations, capsules containing granulations, and pharmaceutical tablets compressed from such granules. In these formulation there is employed from 1% to 85% by weight of the pharmaceutically active agent and its salt form, from 5% to 99% of hydrolyzed cellulose, based on the dry weight of the granulation, and optionally, conventional granulation and/or tableting additives such as surfactants, disintegrants, and antiadherents/flow aids. Said tablets have singificantly increased dissolution of the pharmaceutically active agent at the pH of the gastorintestinal tract in comparison with the unconverted free pharmaceutically active agent.

L'invention concerne un procédé de préparation d'une formulation granulaire compressible séchée par atomisation, destiné à préparer des comprimés pharmaceutiques. Selon le procédé, des agents pharmaceutiquement actifs essentiellement insolubles à l'eau, acides, amphotères et basiques sont transformés en sels davantage solubles à l'eau, qui sont réduits en grains à l'aide de cellulose hydrolysée. L'invention concerne également des coulis contenant des médicaments, les granulations obtenues, des capsules contenant les granulations et des comprimés pharmaceutiques comprimés à partir de ces granules. Dans ces formulations, sont utilisés de 1 à 85 % en pds de l'agent pharmaceutiquement actif et son sel, de 5 à 99 % de cellulose hydrolysée, sur la base du poids sec de la granulation, et éventuellement, une granulation habituelle et/ou des additifs de fabrication de comprimés, tels que des tensioactifs, des désintégrants et des adjuvants facilant l'écoulement ou l'antiadhérence. Ces comprimés ont un meilleur pouvoir de dissolution de l'agent pharmaceutiquement actif au pH du tractus gastro-intestinal, comparé à l'agent libre pharmaceutiquement actif non transformé.
