A I-CreI homing endonuclease variant obtainable by said method, a vector encoding said variant, a cell, an animal or a plant modified by said vector. Use of said I- CreI endonuclease variant and derived products for genetic engineering, genome therapy and antiviral therapy.
L'invention concerne une méthode permettant de mettre au point par génie génétique des variants de l'endonucléase Homing I-CreI capables de cliver des sites mutants I-CreI présentant une variation aux positions ± 8 à ±10. L'invention concerne également un variant de l'endonucléase Homing I-CreI obtenu au moyen de cette méthode, un vecteur codant pour ledit variant, une cellule, un animal ou une plante modifié au moyen dudit vecteur. L'invention concerne enfin l'utilisation du variant de l'endonucléase Homing I-CreI et de produits dérivés pour le génie génétique, la thérapie du génome et la thérapie antivirale.
