A group of bacterial dihydroxy-acid dehydratases having a [2Fe-2S] cluster was discovered. Bacterial [2Fe-2S] DHADs were expressed as heterologous proteins in bacteria and yeast cells, providing DHAD activity for conversion of 2,3-dihy- droxyisovalerate to .alpha.-ketoisovalerate or 2,3-dihydroxymethylvalerate to .alpha.-ketomethylvalerate. Isobutanol and other compounds may be synthesized in pathways that include bacterial [2Fe-2S] DHAD activity.

Linvention concerne un groupe de dihydroxyacide déshydratases bactériennes possédant un agrégat [2Fe-2S]. Des DHAD [2Fe-2S] bactériennes ont été exprimées en tant que protéines hétérologues dans des bactéries et des cellules de levure, fournissant une activité de DHAD pour la conversion de 2,3-dihydroxyisovalérate en a-cétoisovalérate ou de 2,3-dihydroxyméthylvalérate en a-cétométhylvalérate.
