The present invention relates, first, to the discovery, identification, and characterization of novel nucleic acid molecules, that are associated with PBC, Sjögren's syndrome, scleroderma, SLE, autoimmune thyroiditis and various other connective tissue disorders. The novel nucleotide sequences of the present invention are retroviral in origin and are indicative of a PBC retrovirus which bears a strong correlation with PBC. The present invention is based, in part, on the Applicant's data which is the first evidence to suggest that PBC patient's tissue may harbor a transmissible agent. The association of a retroviral infectious agent with PBC was first demonstrated by Applicants <i>in vitro</i> by co-culture of periportal lymph nodes derived from patients at time of transplantation and healthy biliary epithelium cells. The Applicant's discoveries as described herein, report the characterization of PBC-associated infectious agent as retroviral as demonstrated by electron microscopy and immunoblot reactivity. In addition, Applicants have characterized novel nucleotide sequences which are associated with the PBC- associated retrovirus.

L'invention porte premièrement sur la découverte, l'identification et la caractérisation de nouvelles molécules associées à la PBC, au syndrome de Sjögren's, à la sclérodermie, au SLE (lupus érythémateux aigu disséminé), à la thyroïdite auto-immune, et à divers autres troubles des tissus conjonctifs. Ces nouvelles séquences de nucléotides sont d'origine rétrovirale et indicatrices de la présence d'un rétrovirus de la PBC présentant une forte corrélation avec la PBC. Ladite invention se fonde en partie sur des données des déposants, premières constatations suggérant que les tissus d'un patient atteint de PBC pouvaient abriter un agent transmissible. L'association d'agents infectieux rétroviraux à la PBC a d'abord été démontrée <i>in vitro</i> par les déposants par une co-culture de ganglions lymphatiques périportes prélevés sur le patient lors de la transplantation, et de cellules saines d'épithélium biliaire. Les découvertes des déposants exposées dans la description mentionnent que l'agent infectieux associé à la PBC à un caractère rétroviral, comme cela a été démontré par le microscope électronique et la réactivité de l'immunoblot. Les déposants ont en outre caractérisé de nouvelles séquences nucléotidiques associées au rétrovirus associé à la PBC.
