A method for identifying a gene having a role in the presentation of diabetic nephropathy comprises culturing mesangial cells in the presence of a concentration of glucose sufficient to induce differential expression, especially up-regulation, of a gene susceptible to such differential expression and identifying the gene so induced. The cells are also optionally subjected to mechanical strain and/or TGF-.beta.1 can be added to the culture medium. The differentially expressed genes can be identified by suppression subtractive hybridisation. The method has resulted in the identification of novel genes which play a role in the presentation of diabetic nephropathy. The genes can be used as diagnostic markers for diabetic nephropathy and as the basis of drug development programmes.
Cette invention concerne un procédé qui permet d'identifier un gène jouant un rôle dans la présence de néphropathie diabétique. Ce procédé consiste à cultiver des cellules mésangiales en présence d'une concentration de glucose suffisante pour induire une expression différentielle, notamment une régulation vers le haut, d'un gène sensible à une telle expression différentielle, puis à identifier le gène ainsi exprimé. Les cellules peuvent également être soumises à des contraintes mécaniques et/ou un TGF-.beta.1 peut être ajouté au milieu de culture. Les gènes exprimés différentiellement peuvent être identifiés par hybridation soustractive de suppression. Ce procédé a permis d'identifier de nouveaux gènes qui jouent un rôle dans la présence de néphropathie diabétique. Ces gènes peuvent être utilisés en qualité de marqueurs diagnostiques de néphropathie diabétique, et peuvent servir de base pour des programmes d'élaboration de médicaments.
