A recombinant DNA molecule coding for a protein expressed by a Staphylococcus aureus bacterium, comprising the nucleotide sequence SEQ ID NO: 1 or a homologous sequence, or a partial or homologous sequence of SEQ ID NO: 1 coding for a polypeptide fragment comprising at least 15 amino acid residues, is described. Further, a protein expressed by such a bacterium or a polypeptide fragment comprising at least 15 amino acid residues, comprising the amino acid sequence SEQ ID NO: 2 binds IgG and apolipoprotein H. Examples of the polypeptide fragments comprise the SEQ ID NO: 3 through 6. These proteins and polypeptide fragments may be coupled to an inert carrier or matrix. Vectors comprising such a DNA molecule or the corresponding RNA molecule, and antibodies specifically binding to a polypeptide having an amino acid sequence of SEQ ID NO:4 or SEQ ID NO:6, are also disclosed. The DNA or RNA molecules, the vectors and the antibodies mentioned may all be used in different types of vaccines against Staphylococcal infections. Moreover, a method of isolating and/or purifying apoliprotein H from a liquid medium, especially from serum, is described.
Cette invention a trait à une molécule d'ADN de recombinaison codant une protéine exprimée par Staphylococcus aureus, comprenant la séquence nucléotidique SEQ ID N· 1 ou une séquence homologue ou une séquence partielle ou homologue de SEQ ID N· 1 codant un fragment de polypeptide comportant au moins 15 résidus d'aminoacide. Elle porte également sur une protéine exprimée par cette bactérie ou par un fragment de polypeptide comportant au moins 15 résidus d'aminoacide, comprenant la séquence aminoacide SEQ ID N· 2. La protéine en rapport avec la SEQ ID N· 2 fixe l'IgG et l'apolipoprotéine H. Des exemples de ces fragments de polypeptides comportent les séquences SEQ ID N· 3 à N· 6. Ces protéines et ces fragments de polypeptide peuvent être couplés à un porteur ou à une matrice inertes. Cette invention concerne également la molécule d'ADN susmentionnée ou une molécule correspondante d'ARN ainsi que des anticorps se fixant de manière spécifique à un polypeptide possédant une séquence aminoacide SEQ ID N· 4 ou N· 6. Les molécules d'ADN ou d'ARN ainsi que les vecteurs et les anticorps mentionnés peuvent être utilisés dans différents types de vaccins contre des infections à staphylocoque. Il est également traité d'une méthode permettant d'isoler et/ou de purifier l'apolipoprotéine H issue d'un milieu liquide, de sérum notamment.
