Abstract of the Disclosure An antibody immobilized on a water-insoluble solid support by a heterobifunctional cross-linking reagent, wherein said antibody is a partially reduced form of immunogloblin being specific to human proinsulin C-peptide and having thiol groups, is disclosed. The solid support has amino groups and the cross-linking reagent is capable of forming two bonds, one with the thiol group and the other with the amino group. The antibody is useful in conducting immunoassay for proinsulin C-peptide in body fluid. Purification of an antiserum specific to human proinsulin C-peptide by means of immunoaffinity chromatography on 2 column carrying human proinsulin C-peptide or its fragment as ligand and the subsequent partial reduction and immobilization to a water-insoluble solid support with the aid of the heterobifunctional cross-linking reagent afford an excellent immobilized antibody which shows higher specificity and sensitivity than the original antiserum. A radioimmunoassay kit for the measurement of human proinsulin C-peptide in body fluid comprising said immobilized antibody, standard human proinsulin C-peptide solution, a radioactively labelled derivative of human proinsulin C-peptide and a buffer is also disclosed.
