The present invention is based on the finding that antibodies raised against a fragment of PAR1-released peptide may be used to detect in a bodily fluid sample from a subject a marker associated with cancer state, if said subject has cancer. Thus, the present invention provides the methods and packages for conducting one or more of the following: determining a cancerous state in a subject, the method comprises determining binding of an antibody raised against a protease-activated receptor 1 (PAR1) released peptide or a fragment derived therefrom to a marker within a fluid sample obtained from said subject, wherein binding of said antibody to said marker being indicative of a cancerous state; determining severity of a cancerous state in a subject comprising determining level of binding of an antibody raised against PAR1 released peptide or a fragment derived therefrom to a marker within a fluid sample obtained from said subject, and comparing the level of binding with the level of prior determined standards that correlate level of antibody binding to PAR1 released peptide with severity of cancerous state; and determining the effectiveness of a therapeutic treatment of a subject with an anti-cancer agent to the subject comprising determining the level of binding of an antibody raised against PAR1 released peptide or a fragment derived therefrom to a marker within a fluid sample obtained from said subject in two or more successive time points, one or more time points are during the therapeutic treatment, wherein a difference in the level being indicative of effectiveness of therapeutic treatment.
La présente invention se base sur la découverte selon laquelle des anticorps dirigés contre un fragment de peptide libéré par PAR1 peuvent être utilisés pour détecter dans un échantillon de fluide corporel provenant d'un sujet un marqueur associé à un état cancéreux, si ledit sujet est atteint du cancer. Par conséquent, la présente invention concerne des procédés et des kits permettant d'opérer une ou plusieurs des déterminations suivantes : détermination d'un état cancéreux chez un sujet, le procédé comprenant la détermination de la liaison d'un anticorps dirigé contre un peptide libéré par un récepteur 1 activé par une protéase (PAR1) ou contre un fragment dérivé de celui-ci à un marqueur présent dans un fluide corporel obtenu dudit sujet, la liaison dudit anticorps audit marqueur étant indicatrice d'un état cancéreux; la détermination de la sévérité d'un état cancéreux chez un sujet comprenant la détermination du niveau de liaison d'un anticorps dirigé contre un peptide libéré par PAR1 ou contre un fragment dérivé de celui-ci à un marqueur présent dans un fluide corporel obtenu dudit sujet, et la comparaison du niveau de liaison à des niveaux de référence déterminés antérieurement qui corrèlent le niveau de liaison de l'anticorps au peptide libéré par PAR1 avec la sévérité de l'état cancéreux; et la détermination de l'efficacité du traitement thérapeutique d'un sujet avec un agent anticancéreux administré au sujet comprenant la détermination du niveau de liaison d'un anticorps dirigé contre un peptide libéré par PAR1 ou contre un fragment dérivé de celui-ci à un marqueur présent dans un fluide corporel obtenu dudit sujet en deux points temporels successifs ou plus, un ou plusieurs des points temporels se situant pendant le traitement thérapeutique et toute différence de niveau étant indicatrice de l'efficacité du traitement thérapeutique.
