The present invention relates to a monoclonal antibody 20D3 which specifically recognizes the RNA:DNA hybrid (HNA) and does not cross-react with single- stranded or double-stranded DNA or RNA. The present invention also relates to an ELISA method for the immunodetection of DNA:RNA hybrid (HNA) to indicate the absence or presence of a pathogen in a sample, which comprises the steps of: a) incubating said sample with at least one DNA probe specific to said pathogen RNA for a time sufficient for HNA formation to occur; b) incubating said HNA of step a) with the monoclonal antibody of the present invention for antibody-HNA duplex to occur; c) detecting said antibody-HNA duplex of step b); thereby determining the presence of said pathogen in said sample, such as a Listeria sp. or an oncogene expressed by a tumor.
