It is intended to provide an immunogen and a composition for immunological use by which a sufficient immune response can be induced even from an antigen protein seemingly unavailable to induce any effective immune responses for various reasons, and a process for producing an antibody using the same. An immunogen containing a fused protein in which the full-length or a part of a desired antigen protein and a folding factor or its subunit are bonded together via one or more peptide bonds. Owing to the function of the folding factor, the antigen protein contained in the fused protein can be obtained as a soluble protein having a correct stereostructure. It is also possible to prevent the antigen protein from quick degradation in the blood of an animal having been inoculated therewith. Compared with a single antigen protein, furthermore, this immunogen can induce an elevated immune reaction in an animal. Examples of the folding factor include chaperonine and PPIase.
L'invention concerne un agent immunogène et une composition destinée à une utilisation immunologique où une réaction immunitaire suffisante peut être induite, même à partir d'une protéine antigénique apparemment non apte à induire une quelconque réaction immunitaire suffisante pour différentes raisons. La présente invention porte également sur un procédé pour produire un anticorps au moyen de ces éléments. Selon l'invention, un agent immunogène contient une protéine fusionnée, dans laquelle la longueur totale ou partielle d'une protéine antigénique sélectionnée et un facteur d'assemblage ou une sous-unité sont liés ensemble par une ou plusieurs liaisons peptidiques. Vu la fonction du facteur d'assemblage, la protéine antigénique contenue dans la protéine fusionnée peut être obtenue sous forme de protéine soluble dont la stéréostructure est correcte. Il est également possible d'éviter la dégradation rapide de la protéine antigénique dans le sang d'un animal inoculé. Comparativement à une protéine antigénique isolée, ledit agent immunogène peut en outre induire une forte réaction immunitaire chez un animal. Des exemples du facteur d'assemblage comprennent la chaperonine et la PPIase.
