A vaccine composition comprising a Th1-inducing adjuvant in combination with a protecting Liver Stage Antigen or immunological fragment thereof of a human malaria parasite, especially Plasmodium falciparum, with the proviso that when the immunological fragment is an immunological fragment of LSA-3 the Th1- inducing adjuvant is not Montanide. In one preferred aspect the Th1-inducing adjuvant comprises QS21, De-O-acylated monophosphoryl lipid A (3D-MPL) and an oil in water emulsion wherein the oil in water emulsion has the following composition: a metabolisible oil, such a squalene, alpha tocopherol and tween 80. In a further preferred aspect the protecting Liver Stage Antigen is Liver Stage Antigen 3 (LSA-3) or an immunological fragment thereof. A multivalent vaccine composition is also provided comprising the vaccine composition of the invention and in addition at least one other protecting antigen or an immunological fragment thereof, of a malaria parasite.
La présente invention concerne une composition vaccinale comprenant un adjuvant induisant la réponse des cellules Th1 en combinaison avec un antigène protecteur spécifique du foie, ou un fragment immunologique dudit antigène, d'un parasite du paludisme humain, en particulier le Plasmodium falciparum, à la condition que lorsque le fragment immunologique est un fragment immunologique de LSA-3, l'adjuvant induisant la réponse des cellules Th1 n'est pas du Montanide. Dans un mode de réalisation préféré, l'adjuvant induisant la réponse des cellules Th1 comprend de la QS21, un lipide A de monophosphoryle 3-De-O-acétylé (3D-MPL) et une émulsion d'huile dans l'eau, laquelle présente la composition suivante: une huile pouvant être métabolisée, telle qu'un squalène, de la vitamine E et une tween 80. Dans un autre mode de réalisation préféré, l'antigène protecteur spécifique du foie est l'antigène spécifique du foie 3 (LSA-3) ou un fragment immunologique dudit antigène. L'invention concerne également une composition vaccinale polyvalente comprenant la composition vaccinale de l'invention et au moins un autre antigène protecteur ou un fragment immunologique dudit antigène, d'un parasite paludéen.
