An antisense oligonucleotide compound, composition, vaccine and methods for treating a variety of conditions characterized by up-regulation of IL-10 in a mammalian subject are disclosed. The compound (i) is composed of morpholino subunits and phosphorus-containing intersubunit linkages joining a morpholino nitrogen of one subunit to a 5' exocyclic carbon of an adjacent subunit, (ii) is capable of uptake by monocytes, lymphocytes, and dendritic cells in a mammalian subject, (iii) contains between 10-40 nucleotide bases, and (iv) has a base sequence effective to hybridize to at least 12 contiguous bases of a target sequence contained in an exon-2 or exon-4 slice site region of human IL-10 pre- mRNA.

L'invention se rapporte à un composé oligonucléotide antisens, une composition, un vaccin et méthodes destinés au traitement d'une variété d'états caractérisée par la régulation à la hausse d'IL-10 chez un sujet mammifère. Le composé (i) est constitué de sous-unités morpholino et de liaisons entre sous-unités contenant du phosphore, joignant un azote morpholino d'une sous-unité à un carbone exocyclique 5' d'une sous-unité adjacente, (ii) est apte au recaptage par des cellules monocytes, lymphocytes et dendritiques chez un sujet mammifère, (iii) contient entre 10 et 40 bases nucléotides, et (iv) possède une séquence de base efficace pour hybrider jusqu'à au moins 12 bases contigües d'une séquence cible contenue dans une région de mutation délétère exon-2 ou exon-4 d'un IL-10 pre-mARN humain.
