A method is provided for enhancing the reactivity of a T cell toward a tumour cell which method comprises: (a) isolating a T cell which is a tumour infiltrating lymphocyte (TIL) from a patient having said tumour cell present in their body; (b) introducing a nucleic acid sequence into the TIL, which sequence is capable of inhibiting or preventing expression of an endogenous Notch ligand in the TIL; and (c) re-introducing the transfected TIL into the patient; wherein the T cell comprises a T cell receptor specific for a tumour antigen expressed by the tumour cell.
La présente invention concerne un procédé servant à améliorer la réactivité d'une cellule T à une cellule tumorale, ledit procédé comprenant les étapes suivantes: (a) isolation d'une cellule T qui est un lymphocyte d'infiltration tumorale (tumour infiltrating lymphocyte/ TIL), chez un patient ayant ladite cellule tumorale présente dans son corps; (b) introduction d'une séquence d'acide nucléique dans le TIL, ladite séquence étant capable d'inhiber ou de bloquer l'expression d'un ligand de Notch endogène dans le TIL; et (c) réintroduction du TIL transfecté chez le patient. Selon cette invention, la cellule T comprend un récepteur de cellule T spécifique d'un antigène tumoral exprimé par la cellule tumorale.
