Described is a method for the preparation of crosslinked enzyme aggregates, comprising enzyme molecules being associated with an enzyme activity-enhancing agent, the enzyme activity-enhancing agent interacting with the said enzyme molecules, comprising the steps of: A - providing a plurality of enzyme molecules, B - aggregating the enzyme molecules in a liquid medium, by means of addition of a precipitating agent, C - crosslinking the aggregated enzyme molecules to one another by providing a crosslinking agent in the liquid medium, the method comprising, during or between steps A, B or C, the step of adding an enzyme activity-enhancing agent and allowing the enzyme molecules to become and to remain associated with the enzyme activity-enhancing agent. The resulting CLEA are advantageous due to increase in activity, their suitability for recycle, enhanced stability and low requirements for additional enzyme activity-enhancing agent. Further, crosslinked enzyme aggregates are described, comprising an enzyme activity-enhancing agent associated therewith.

Cette invention concerne un procédé de préparation d'agrégats d'enzymes réticulées comprenant des molécules enzymatiques associées à un agent facilitant l'activité enzymatique, lequel agent facilitant l'activité enzymatique interagit avec lesdites molécules enzymatiques. Ce procédé comprend les étapes consistant : (A) à utiliser une pluralité de molécules enzymatiques ; (B) à agréger les molécules enzymatiques dans un milieu liquide au moyen d'un agent de précipitation ajouté audit milieu liquide ; et (C) à réticuler les molécules enzymatiques agrégées les unes avec les autres au moyen d'un agent de réticulation ajouté audit milieu liquide. Ce procédé comprend, pendant ou entre les étapes A, B ou C, une étape consistant à ajouter un agent facilitant l'activité enzymatique et à permettre aux molécules enzymatiques de s'associer et de rester associées à l'agent facilitant l'activité enzymatique. Les agrégats d'enzymes réticulées ainsi obtenus ont l'avantage de présenter une plus forte activité, une aptitude au recyclage, une plus grande stabilité et un faible besoin en agent facilitant l'activité enzymatique supplémentaire. Cette invention concerne en outre des agrégats d'enzymes réticulées comprenant un agent facilitant l'activité enzymatique associé auxdits agrégats.
