The present invention relates to improved methods to immunize/vaccinate or stimulate the immune system of animals, including humans, using vectors containing expression cassettes that encode for the DNA of one or more protein/peptide antigens and/or adjuvants, in particular, cytokines like GMCSF, F1t3L, interleukins, and the like, which can be encoded by DNA as well, also recombinase mediated integration. Adjuvants known to increase immune responses following DNA vaccination. In addition, the vectors contain one or more sites recognized by a recombinase/transposase, which catalyzes the insertion of the vector into the genome of transfected cells. Stable integration of the plasmid vector into the genome of transfected cells results in higher and longer-lasting expression of the encoded protein(s), and increases the immune response in the vaccinated animal. The present invention also relates to adjuvant compositions comprising the novel polypeptide, rabbit GMCSF, for boosting antibody production in rabbits.
La présente invention a trait à des procédés améliorés d'immunisation/vaccination ou de stimulation du système immunitaire d'animaux, comprenant les humains, mettant en oeuvre des vecteurs contenant des cassettes d'expression codant pour l'ADN d'un ou de plusieurs antigènes et/ou adjuvants protéiques/peptidiques, notamment, des cytokines telles que GMCSF, F1t3L, des interleukines et analogues, qui peuvent être codées par l'ADN également, et l'intégration liée à la recombinase; des adjuvants réputés pour l'accroissement des réponses immunitaires suite à la vaccination par l'ADN. En outre, les vecteurs contiennent un ou plusieurs sites reconnus par une recombinase/transposase, qui catalyse l'insertion du vecteur dans le génome de cellules transfectées. Une intégration stable du vecteur plasmidique dans le génome de cellules transfectées entraîne une expression plus élevée et de plus longue durée de la/des protéine(s) codée(s), et augmente la réponse immunitaire chez l'animal vacciné. La présente invention a également trait à des compositions d'adjuvant comportant le nouveau polypeptide, la GMCSF de lapin, pour l'amplification de la production d'anticorps chez les lapins.
