The invention provides fibronectin type III (Fn3)-based binding molecules that bind to a specific target antigen. The invention further provides bispecific Fn3 -based binding molecules that bind to two or more targets simultaneously. The Fn3-based binding molecules of the invention can also be linked together to form multispecific Fn3-based binding molecules, and/or can be conjugated to a non-Fn3 moiety, such as, Human Serum Albumin (HSA), for improved half life and stability. The invention also provides methods for generating, screening and using Fn3 -based binding molecules in a variety of therapeutic and diagnostic applications.

La présente invention concerne des molécules de liaison à base de fibronectine de type III (Fn3) qui se lient à un antigène cible spécifique. Linvention concerne en outre des molécules de liaison à base de Fn3 bispécifiques qui se lient simultanément à deux cibles ou plus. Les molécules de liaison à base de Fn3 de linvention peuvent également être liées les unes aux autres pour former des molécules de liaison à base de Fn3 multispécifiques, et/ou peuvent être conjuguées à une fraction non Fn3, telle que la sérum albumine humaine (HSA), pour une meilleure demi-vie et une meilleure stabilité. Linvention concerne également des procédés de génération, de criblage et dutilisation de molécules de liaison à base de Fn3 dans une variété dapplications thérapeutiques et diagnostiques.
