The present invention relates to the treatment of skin defects by organotypically-cultured autologous keratinocytes isolated from the outer root sheath of anagen or growing hair. Methods for primary, as well as subsequent organotypic cultures (i.e., epidermal equivalents) in fully-defined media supplemented by autologous human serum and substances isolated form blood components, with minimal allogeneic biological supplements, are disclosed herein. Techniques to prepare epidermal equivalents for transplantation by use of a biocompatible glue are also disclosed herein.

La présente invention concerne le traitement de lésions cutanées au moyen de kératinocytes autologues cultivés de manière organotypique et isolés de la gaine épithéliale externe des poils en développement ou en phase anagène. L'invention concerne également des méthodes destinées à des cultures organotypiques aussi bien primaires que secondaires (notamment d'équivalents épidermiques) dans des supports entièrement définis complétés par un sérum humain autologue et par des substances isolées de composants sanguins, avec un minimum de compléments biologiques allogéniques. L'invention se rapporte enfin à des techniques de préparation d'équivalents épidermiques destinés à une greffe faisant intervenir une colle biocompatible.
